I noticed the pinion seal on my rear axle was leaking a bit so after i got home from a drive i pulled the rear plug and and a little air hissed out. Could the vent the stuck closed? It looks like a tube with a cap on with slots around the edge is that it, or is there more to it?

Yep, probably stuck or plugged with crap.
When I rebuilt a '74 D30 before installing it in my '71, the drilled hole in the diff casing and vent connection were completely plugged with sand... it was like mortar...
Take off the diff cover, then remove the cap, clean the hole. Install a new connector and run vent lines. :)

loosen it up with water? then vacuum it? that's what i did.

check your flex tube, see if that's plugged. might not be at the connector.
